Tarlatamab is a bispecific T-cell engager (BiTE) antibody that bridges delta-like ligand 3 (DLL3)-expressing tumor cells with CD3-positive T cells, inducing T-cell-mediated cytotoxicity. It is approved for the treatment of extensive-stage small cell lung cancer after prior platinum-based chemotherapy, and is under investigation for bladder cancer and melanoma. DLL3 is highly expressed in small cell lung cancer and other neuroendocrine tumors, making it a highly selective therapeutic target.
