Raltitrexed is a quinazoline folate analog that directly and specifically inhibits thymidylate synthase (TS) after intracellular polyglutamation, thereby depleting thymidine monophosphate and disrupting DNA synthesis and repair in rapidly dividing cells. It is used for the treatment of advanced colorectal cancer and has been investigated in mesothelioma and other malignancies.
