Riociguat is a soluble guanylate cyclase (sGC) stimulator that acts by two distinct mechanisms: sensitizing sGC to endogenous nitric oxide (NO) and directly stimulating sGC independently of NO, thereby increasing cyclic GMP production in vascular smooth muscle and causing vasodilation and antiproliferative effects. It is approved for the treatment of pulmonary arterial hypertension and chronic thromboembolic pulmonary hypertension.
