Tagraxofusp
Tagraxofusp is a CD123-directed cytotoxin consisting of recombinant human interleukin-3 (IL-3) fused to a truncated diphtheria toxin fragment, targeting CD123 (the IL-3 receptor alpha subunit) overexpressed on blastic plasmacytoid dendritic cell neoplasm (BPDCN) cells and AML blasts. After IL-3-mediated CD123 binding and cellular internalization, the diphtheria toxin moiety inhibits protein synthesis by ADP-ribosylating elongation factor 2 (EF-2), causing cell death. It is the first therapy approved specifically for BPDCN.
