Volanesorsen sodium is an antisense oligonucleotide that targets and degrades apolipoprotein C-III (apoC-III) mRNA in hepatocytes via RNase H-mediated cleavage, thereby reducing plasma apoC-III levels that normally inhibit lipoprotein lipase and VLDL clearance. It is approved in Europe for familial chylomicronemia syndrome (FCS), a rare condition of severe hypertriglyceridemia due to lipoprotein lipase pathway defects, reducing triglyceride levels and pancreatitis risk. Its subcutaneous administration provides sustained apoC-III suppression.
