Omeprazole can modestly increase the rate of acetaminophen absorption by altering gastric pH, but the effect on overall bioavailability and therapeutic outcome is not clinically meaningful under most circumstances.
Mecanismo
Omeprazole raises intragastric pH, which can accelerate gastric emptying and modify dissolution of acetaminophen tablets; CYP2E1 inhibition by omeprazole may also slightly reduce NAPQI formation.
Relevancia clínica
The interaction is generally considered clinically insignificant; no cases of acetaminophen toxicity attributable solely to omeprazole co-administration have been reported.
Manejo clínico
No dose adjustment or monitoring is needed; the combination is used routinely and safely.
