Vosoritide is a modified C-type natriuretic peptide (CNP) analog that activates natriuretic peptide receptor 2 (NPR-B) in the growth plate of bones, stimulating endochondral bone formation and counteracting the excessive FGF receptor 3 (FGFR3) signaling responsible for impaired chondrocyte differentiation in achondroplasia. It is approved for the treatment of achondroplasia in pediatric patients aged 2 years and older who have open epiphyses, increasing linear bone growth velocity. Its mechanism directly addresses the molecular pathology of the most common form of disproportionate dwarfism.
