## Overview
Atorvastatin and lovastatin represent a large potency gap within the statin class. Lovastatin, a natural fermentation product from Aspergillus terreus, was among the first statins approved. Atorvastatin, a fully synthetic compound, is roughly 3–5 times more potent per milligram.
## Mechanism of Action
Both inhibit HMG-CoA reductase. Lovastatin is a prodrug requiring lactonase hydrolysis in vivo. Atorvastatin is administered as the active acid. Both are lipophilic and enter hepatocytes via passive diffusion in addition to transporter-mediated uptake.
