Zolbetuximab
Zolbetuximab is a chimeric monoclonal antibody targeting Claudin 18.2 (CLDN18.2), a tight junction protein selectively expressed on gastric and gastroesophageal junction tumor cells, that mediates antitumor activity through antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC) upon CLDN18.2 binding. It is approved in combination with chemotherapy for first-line treatment of HER2-negative, CLDN18.2-positive locally advanced or metastatic gastric or gastroesophageal junction adenocarcinoma. Its targetable tumor-selective expression makes CLDN18.2 a novel antigen in upper gastrointestinal malignancies.
