Tisotumab vedotin is an antibody-drug conjugate (ADC) consisting of a human monoclonal antibody targeting tissue factor (TF/CD142), conjugated via a protease-cleavable linker to monomethyl auristatin E (MMAE), a microtubule-disrupting cytotoxin. Upon binding to TF-expressing tumor cells, the ADC is internalized and MMAE is released, inducing cell cycle arrest and apoptosis; TF-expressing tumor cells also receive direct antibody-mediated cytotoxicity. It is approved for recurrent or metastatic cervical cancer and is under investigation for bladder and non-small cell lung cancers.
