Tafasitamab is a humanized anti-CD19 monoclonal antibody with enhanced Fc-region modifications that increase binding affinity to Fc-gamma receptors and complement proteins, augmenting antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP) against CD19-positive B-cell malignancies. It is approved in combination with lenalidomide for relapsed/refractory diffuse large B-cell lymphoma (DLBCL) and is under investigation for other B-cell lymphomas and solid tumors. CD19 expression across B-cell development makes it a broadly applicable target in B-cell lymphomas.
