Zanidatamab is a bispecific monoclonal antibody targeting two non-overlapping epitopes on the HER2 receptor (human epidermal growth factor receptor 2), providing dual HER2 blockade that inhibits HER2 dimerization, activates antibody-dependent cellular cytotoxicity (ADCC), and increases receptor internalization and degradation more effectively than single-epitope anti-HER2 agents. It is under investigation for HER2-amplified or HER2-overexpressing cancers including gastroesophageal adenocarcinoma and biliary tract cancer.
