Mechanismus
Bupropion is a moderate-to-potent CYP2D6 inhibitor; venlafaxine is extensively metabolized by CYP2D6 to its active O-desmethylvenlafaxine metabolite, resulting in increased venlafaxine exposure.
Klinische Bedeutung
Clinical studies show increased venlafaxine AUC and Cmax with concurrent bupropion; additive noradrenergic effects may raise blood pressure.
Maßnahmen
Monitor blood pressure and serotonin syndrome symptoms; consider venlafaxine dose reduction when adding bupropion.
