Zimeldine was the first selective serotonin reuptake inhibitor (SSRI) to reach clinical use, blocking the serotonin transporter (SERT) to inhibit synaptic serotonin reuptake and increase serotonergic neurotransmission for antidepressant effect. It was the prototype SSRI that established the therapeutic validity of selective serotonin reuptake inhibition. It was withdrawn worldwide due to serious hypersensitivity reactions including Guillain-Barré syndrome, but it paved the way for the development of subsequent SSRIs.
