OAT1 (OAT1)
OAT1Organic Anion Transporter 1 (gene: SLC22A6), a renal uptake transporter expressed on the basolateral membrane of proximal tubule cells. Mediates the first step of renal secretion by transporting organic anions from peritubular blood into tubular cells via exchange with intracellular dicarboxylates (alpha-ketoglutarate). Key determinant of renal clearance for many anionic drugs, including antivirals, diuretics, and NSAIDs.
Examples
- Probenecid inhibition of OAT1 reducing penicillin renal clearance
- Tenofovir nephrotoxicity linked to OAT1-mediated tubular accumulation
- Cidofovir renal uptake and dose-limiting nephrotoxicity
