Phosphodiesterase (PDE)
PDEA family of enzymes (PDE1–PDE11) that hydrolyze cyclic nucleotides cAMP and cGMP, thereby terminating their second messenger signaling. PDE inhibitors increase intracellular cyclic nucleotide levels to produce vasodilation, bronchodilation, anti-inflammatory, or inotropic effects depending on the isoform targeted.
Examples
- Sildenafil — PDE5 inhibitor (erectile dysfunction, pulmonary hypertension)
- Roflumilast — PDE4 inhibitor (COPD)
- Milrinone — PDE3 inhibitor (acute heart failure)
