Pemigatinib
Fibroblast growth factor receptor alterations define the population treated with pemigatinib, a targeted small-molecule therapy. Where an FGFR gene fusion or rearrangement produces a constitutively active receptor, the resulting signal drives proliferation independently of normal growth-factor control; inhibiting the abnormal protein removes that drive. Treatment therefore depends on molecular characterisation of the tumour rather than on its site of origin alone, the pattern common to genomically selected therapies. Selection by genotype also narrows the eligible population sharply compared with histology-defined indications.
