Cross-Tolerance

Pharmacodynamics

A phenomenon in which tolerance developed to one drug confers reduced sensitivity to another drug that acts through the same or a related receptor system or signaling pathway. Cross-tolerance is clinically relevant for drug switching, as patients tolerant to one agent may require higher-than-expected starting doses of a pharmacologically related drug.

  • Opioid cross-tolerance: morphine tolerance partially extends to hydromorphone and fentanyl
  • Benzodiazepine-alcohol cross-tolerance: shared GABA-A receptor potentiation mechanism
  • Amphetamine-methylphenidate partial cross-tolerance through dopaminergic pathway overlap
