Teplizumab is a humanized anti-CD3 monoclonal antibody that binds to the CD3 epsilon chain of the T-cell receptor complex, inducing partial agonism and selectively modulating autoreactive T cells responsible for pancreatic beta-cell destruction in type 1 diabetes. It is approved to delay the onset of stage 3 type 1 diabetes in at-risk individuals aged 8 years and older, representing the first disease-modifying therapy for type 1 diabetes prevention. By shifting the balance from effector to regulatory T cells, it preserves residual beta-cell function.
