Pregabalin
Used to relieve nerve pain caused by conditions such as diabetic neuropathy, shingles, fibromyalgia, and spinal cord injury, and also used as an add-on treatment to control certain types of seizures in epilepsy. It works by reducing abnormal electrical activity in the nervous system that causes pain and seizures.

Pregabalin potentiates the respiratory depressant and sedative effects of morphine, creating a high-risk combination for overdose.
Pregabalin combined with oxycodone produces synergistic respiratory and CNS depression, substantially elevating overdose risk.
Duloxetine and pregabalin are commonly co-prescribed for neuropathic pain with complementary mechanisms; additive CNS effects including dizziness, somnolence, and cognitive impairment can occur.
Omeprazole does not have a pharmacokinetic interaction with pregabalin since pregabalin is not CYP-metabolised; however, both can cause peripheral oedema through independent mechanisms, producing additive fluid retention.
Combining gabapentin and pregabalin, two alpha-2-delta ligands with similar mechanisms, produces additive CNS depression and dose-dependent adverse effects including respiratory depression, dizziness, and somnolence.
