Orphan Receptor
A receptor identified through genomic or proteomic studies whose endogenous ligand and physiological function remain unknown. Deorphanization — identifying the natural ligand — can reveal new therapeutic targets. Many GPCRs, nuclear receptors, and ion channels were initially classified as orphans.
Örnekler
- GPR55 — putative cannabinoid receptor (deorphanized with LPI)
- RORgamma-t — later identified as target of digoxin-like compounds
- GPR119 — deorphanized with OEA, pursued for type 2 diabetes
