Sutimlimab is a humanized monoclonal IgG4 antibody that selectively targets and inhibits C1s, a serine protease in the classical complement pathway, approved for the treatment of hemolysis in adults with cold agglutinin disease (CAD). By blocking C1s-mediated cleavage of C4 and C2, it prevents classical pathway-driven complement activation on red blood cells by cold agglutinin IgM antibodies, inhibiting complement-mediated hemolysis. It is the first targeted therapy for this rare hemolytic anemia.
