Tezepelumab is a human monoclonal antibody that blocks thymic stromal lymphopoietin (TSLP), an epithelial cytokine at the top of the inflammatory cascade that drives IgE production, eosinophil activation, and Th2 inflammation in asthma and atopic dermatitis. It is approved as add-on maintenance therapy for severe asthma regardless of eosinophil phenotype, making it unique among biologics in treating non-eosinophilic asthma. By blocking upstream TSLP, it broadly suppresses multiple allergic and non-allergic inflammatory pathways.
