Autoinduction
A pharmacokinetic phenomenon in which a drug induces its own metabolism by upregulating the enzymes responsible for its biotransformation. This leads to a time-dependent decrease in plasma drug concentrations despite constant dosing, often requiring dose escalation over weeks. Autoinduction complicates dose optimization and therapeutic drug monitoring.
示例
- Carbamazepine: induces CYP3A4, its own half-life decreases from 36 h to 16-24 h over 2-4 weeks
- Artemisinin: autoinduction of CYP2B6 reduces its own bioavailability during repeated dosing
- Rifampin: partially autoinduces its own metabolism through CYP3A4
