Reversible inhibition of the epidermal growth factor receptor tyrosine kinase is the pharmacology of erlotinib hydrochloride, which prevents receptor autophosphorylation and thereby interrupts downstream proliferative signalling. Tumours carrying activating EGFR mutations depend on that pathway, and it is in such tumours that the agent slows growth. The hydrochloride salt is the form presented in standard oral capsules, chosen for pharmaceutical rather than pharmacological reasons. Reversibility of binding distinguishes this generation of EGFR inhibitors from the covalent agents developed subsequently. The salt has the formula C22H24ClN3O4 and a molecular weight of approximately 429.9 g/mol, and it is approved with development completed through the final phase.

The hydrochloride salt form of erlotinib, this targeted cancer drug blocks the EGFR signaling pathway to slow the growth of EGFR-mutated tumors. The hydrochloride salt formulation is used in standard oral capsules.
